Conjunctival Commensal Isolation and Identification in Mice

The ocular surface was once considered immune privileged and abiotic, but recently it appears that there is a small, persistent commensal presence. Identification monitoring of bacterial species at the mucosa have been challenging due to their low abundance limited availability appropriate methodology for growth identification. There are two standard approaches: culture based or DNA sequencing methods. first method problematic recoverable bacteria second approach identifies both live dead leading an aberrant representation space. We developed robust sensitive isolation by building upon microbiological culturing techniques. This swab-based technique, utilizing “in-lab” made thin swab targets lower conjunctiva, followed amplification step aerobic facultative anaerobic genera. protocol has allowed us isolate identify conjunctival such as Corynebacterium spp., Coagulase Negative Staphylococcus Streptococcus etc. suitable define diversity in mice under different disease conditions.
